What is Isopropylparaben?

Isopropyl 4-hydroxybenzoate is a carboxylic acid ester derivative that has a wide range of applications. It can be used in soy sauce, vinegar, soft drinks (except soda), fruit flavorings, fruits and vegetables, pickled products, and more. Additionally, it is commonly used in the food and cosmetics industries, as well as in pharmaceuticals as an antiseptic, antifungal agent, and fungicide.

Isopropylparaben Uses

  1. Isopropyl 4-hydroxybenzoate, also known as isopropyl paraben, is commonly used in combination with isopropyl ester, butyl ester, and other compounds as an oil-in-water emulsifier. It has a higher solubility compared to pure butyl ester, making it easy to use in soy sauce.
  2. Isopropyl paraben, similar to butyl paraben, can function as a food preservative and antifungal agent. In Japan, six types of parabens are permitted for use, including ethyl, propyl, isopropyl, butyl, isobutyl, and other butyl. Among these, butyl paraben has the strongest antibacterial effect. Generally, parabens with a higher number of carbon atoms in the alcohol component of the ester exhibit stronger antibacterial properties. However, they also tend to have higher toxicity. Isopropyl paraben demonstrates inhibitory or killing effects on various molds and yeasts, and is relatively unaffected by pH. Its use is restricted to soy sauce, vinegar, soft drinks, sherbet, fruit sauce, and fruits and vegetables (limited to the skin part). The recommended dosage is 1.4 times that of parahydroxybenzoic acid.

Isopropylparaben Preparation Method

There are numerous domestic reports detailing the preparation of Isopropylparaben, most of which involve the use of p-hydroxybenzoic acid and isopropyl alcohol in the presence of sulfuric acid. However, the product content and yield of such a synthetic route are very low. This is due to the large steric hindrance of isopropyl alcohol, making it difficult to dehydrate into ester using ordinary catalysts and p-hydroxybenzoic acid. Additionally, this type of reaction is reversible, leading to low synthesis efficiency, low yield, and high production cost.

A new method for the preparation of Isopropylparaben is characterized by the following steps:

A. Isopropyl alcohol is placed into a reaction kettle equipped with a stirring device, a condensing device, and a freezing device, and stirred and frozen.

B. Thionyl chloride is added dropwise to the frozen isopropyl alcohol while stirring continuously and controlling the reaction temperature to remain low. The total amount of thionyl chloride added is: eta isopropyl alcohol: n thionyl chloride = 1:0.2-1:0.3. After the dropwise addition is completed, the freeze is removed, and stirring is continued for 2-3 hours. The chemical equation of the reaction between isopropyl alcohol and thionyl chloride at low temperature is as shown in formula (I).


C. P-hydroxybenzoic acid is added to the reaction kettle, with n isopropyl alcohol: n p-hydroxybenzoic acid = 1:0.18-1:0.20. The mixture is stirred at room temperature for 3-5 hours, slowly heated to reflux, and stirred in the reflux state for 5-6 hours. The reaction is stopped when the sampling HPLC detects that the conversion rate reaches more than 85%. The reaction equation for this step is as shown in formula (II).


D. Pure water is added to the reaction kettle, and the temperature is raised to 80°C. The liquids are separated while hot, and the lower oil is separated. The oil is kept warm and toluene is added to completely dissolve it. The amount of toluene added is eta isopropyl alcohol: n Toluene=1:0. 1-1:0. 12.

E. The solution obtained in step D undergoes crystallization and recrystallization operations until the purity of the crystallized product of isopropyl parahydroxybenzoate is qualified. The qualified product is air-dried below 65°C. The stirring temperature in step A ranges from freezing to no higher than 0°C. The low temperature state described in step B is to keep the reaction temperature no higher than 5°C. The temperature rise rate from slowly heating to reflux in step C is 1°C/min.
