The method for extracting Genivida Genistein from plant Sophora japonica is characterized by comprising the following steps:
(1) Preparation of Genivida Genistein molecularly imprinted polymer:
1) Multi-step swelling polymerization:
a. First, add monodisperse microspheres to microemulsion A, and at room temperature, react with a rotating speed of 100-150 rpm in a constant temperature magnetic stirrer for 14-16 hours to obtain the first-step swelling solution;
The microemulsion A is configured by sodium dodecyl sulfonate, dibutyl phthalate and three distilled water, wherein each milliliter of microemulsion A contains 0.001-0.003 g of sodium dodecyl sulfonate, containing 0.03~0.05g of dibutyl phthalate, and the rest are three distilled water;
b. Add microemulsion B to the first-step swelling solution, and at room temperature, react with a rotating speed of 100-150rpm in a constant temperature magnetic stirrer for 1.5-2.5h to obtain the second-step swelling solution;
The microemulsion B is prepared from an initiator, chloroform, polyvinyl alcohol and three-distilled water, wherein each milliliter of the microemulsion B contains an initiator 0.01-0.02 g, chloroform 0.070-0.100 g and polyvinyl alcohol 0.018~0.025g, the rest are three distilled water;
c. Then add microemulsion C to the swelling solution in the second step, and at room temperature, in a constant temperature magnetic stirrer, react for 1.5-2.5 h at a rotational speed of 100-150 rpm to obtain the swelling solution in the third step;
The microemulsion C is made of genistein, functional monomer, cross-linking agent, cyclohexanol, sodium dodecyl sulfonate, polyvinyl alcohol and three distilled water, wherein each milliliter of microemulsion C contains genistein. 0.008g ~ 0.009g of elemental, functional monomer 0.022g ~ 0.024g, crosslinking agent 0.150 ~ 0.158g, cyclohexanol 0.105 ~ 0.110g, sodium dodecyl sulfonate 0.001 ~ 0.005g and polyvinyl alcohol 0.015 ~ 0.020g g, the rest are three distilled water;
d. Finally, thermally polymerizing the swelling liquid in the third step, specifically: under the protection of nitrogen flow, in a constant temperature magnetic stirrer at a temperature of 50° C. and a rotating speed of 180 rpm for 24 hours to obtain microspheres with a particle size of 1 to 15 μm, So as to complete the swelling polymerization;
2) After the multi-step swelling polymerization is completed, the obtained microspheres are subjected to six times of sedimentation, and the six times of sedimentation are: first, three times in methanol, then once in three-distilled water, and finally twice in tetrahydrofuran. The lower layer of sediment was taken out and filtered with a G4 funnel, and then the filter residue was dried at normal temperature and pressure to obtain genistein molecularly imprinted polymer;
(2) Preparation of Genivida Genistein molecularly imprinted solid phase extraction column:
The genistein molecularly imprinted polymer prepared in step (1) was packed into a column by the homogenization method, and then acetonitrile solution with a volume ratio of 20%, methanol-glacial acetic acid with a volume ratio of 4:1, and a volume ratio of 20% acetonitrile solution for elution to obtain a genistein molecularly imprinted solid phase extraction column;
(3) Genivida Genistein molecularly imprinted solid-phase extraction column to extract and separate genistein from the plant Sophora japonica.
