Dl Malic Acid Uses

Dl Malic Acid Production Method

Preparation of Dl Malic Acid by Microbial Fermentation

There are two types of Dl Malic Acid fermentation processes: one-step fermentation and two-step fermentation. The microorganisms used in different fermentation processes are also different.
(1) The strains of the one-step fermentation method include Aspergillus flavus, Aspergillus oryzae, Aspergillus parasiticus, etc., all of which use various sugars including glucose as substrates to produce Dl Malic Acid.
(2) The two-step fermentation method uses Rhizopus sinensis or Rhizopus aureus to biosynthesize fumaric acid from glucose, and then converts fumaric acid to L – Malic acid. The one-step fermentation method mostly uses Aspergillus flavus, which can produce Dl Malic Acid from various sugary raw materials such as glucose, maltose, sucrose, fructose and starch. However, the medium is complicated, and other organic acids such as fumaric acid are often produced during the fermentation process, which brings difficulties to separation and purification, and it is difficult to obtain high-purity crystals. In addition, the acid production is low, and the conversion rate to sugar is low (30%~40%) %), the cycle is long (at least also need 5 days), so industrialized production has not been realized at home and abroad so far.
In the first step of the two-step fermentation method, rhizopus is used to ferment sugars to produce fumaric acid, that is, fumaric acid fermentation. Glucose is used as the sugar here, and glucose can be obtained by double-enzyme hydrolysis of starch. The second step is to insert yeast or bacteria into the fumaric acid produced above for fermentation, and convert fumaric acid into Dl Malic Acid, that is, conversion fermentation. Using this method, the maximum conversion rate of malic acid to sugar can reach more than 60%, but after all, the fermentation cycle is too long, and it takes 8-9 days to add up. Although the two-step method is the same as the one-step method, scholars at home and abroad are constantly exploring and have made some valuable progress, but there is still no report on industrial production.
Whether it is one-step or two-step fermentation, if starch is used as raw material, it needs to be hydrolyzed before use to make it into glucose, and the best hydrolysis method for starch is double-enzyme hydrolysis.
At present, the methods for industrial production of Dl Malic Acid at home and abroad are all immobilized cell transformation methods. The method is to firstly cultivate a large number of bacteria with high fumarase activity, collect the bacteria and fix them with appropriate carriers such as carrageenan, then put them into a column reactor, use sodium fumarate as a substrate, and flow through The reactor, catalyzed by the fumarase in the immobilized cells, can easily convert fumaric acid into Dl Malic Acid with almost no by-products, and the conversion rate is as high as 95%. Compared with the above two methods, this method is simple, economical and efficient, so it has become the first method for industrial production.
