What is Carboxymethyl Chitosan?

Carboxymethyl chitosan is a derivative of chitosan that is soluble in water and acts as an amphoteric polyelectrolyte. It is produced by adding a hydrophilic group -CH2COOH to the chitosan polymer chain. This modification improves the water solubility of chitosan, especially in neutral and alkaline solutions, and gives it properties such as film-forming, thickening, moisturizing, and chelating. As a result, carboxymethyl chitosan is widely used in agriculture, medicine, health products, cosmetics, preservation, environmental protection, and other industries.

Biomedical Applications of Carboxymethyl Chitosan

(1) Carboxymethyl Chitosan has shown potential in the development of multifunctional wound dressings due to its solubility, antibacterial properties, biodegradability, antioxidant properties, and biocompatibility. Several mechanisms by which carboxymethyl chitosan-based hydrogel materials promote wound healing are summarized, and the use of carboxymethyl chitosan-based hydrogels for wound healing is introduced.

(2) In tissue engineering, Carboxymethyl Chitosan has been used to prepare various biomaterials, including hydrogels, scaffolds, nanoparticles, nanofibers, composites, dendrimers, and membranes. These materials are utilized in the fields of bone/cartilage and organ tissue engineering, taking advantage of the processability of CMC for functionalization and application.

(3) Carboxymethyl Chitosan derivatives are utilized as carriers for a variety of bioactive substances in drug delivery, including anti-inflammatory, antibacterial, anticancer, and antifungal drugs, as well as proteins, peptides, DNA, and genes. Hydrogels, polyelectrolyte complexes, biodegradable release systems, and drug adjuvants are the main platforms for drug delivery using CMC derivatives.

(4) In biological imaging, researchers combine CMC derivatives with other organic molecules, inorganic semiconductors, and substances for non-invasive study of tissue structures and physiological functions. The imaging system is highly selective, fast, and ultra-sensitive, showing promise for the development of biological imaging.

(5) Water-soluble CS derivatives have potential applications in the cosmetics industry, serving as hygroscopic or water-retaining agents, emulsion stabilizing materials, antioxidants, or antibacterials. CS and its derivatives can adhere to the skin and provide moisturizing effects due to their relatively high molecular weight and positive charge.

Purification Method of Carboxymethyl Chitosan

The process of purifying carboxymethyl chitosan involves several steps:

1) Dissolve 40Kg of carboxymethyl chitosan with an isoelectric point pH of 2.7, a number average molecular weight of 40W, and a carboxymethyl substitution degree of 1.3 in 2 tons of water. Adjust the pH of the carboxymethyl chitosan aqueous solution to 2.7, leading to the formation of a white flocculent precipitate. Centrifuge and dehydrate the precipitate to obtain concentrated carboxymethyl chitosan.

2) Wash the concentrated carboxymethyl chitosan with filtered water at pH 2.7 to remove salt and obtain desalted carboxymethyl chitosan.

3) Repeatedly wash the desalted carboxymethyl chitosan with 95% ethanol until the alcohol content of the supernatant is 95°. Remove the supernatant, centrifuge, and dry to obtain carboxymethyl chitosan with a purity of 99.3% sugar.

This purification process used a total of 430Kg of 95% ethanol. Compared to existing technology, which requires 2 tons of 95% ethanol to purify 40Kg of the same carboxymethyl chitosan, this method saves a significant amount of ethanol, reducing costs and environmental impact.

Carboxymethyl Chitosan Preparation

The sodium hydroxide solution in isopropyl alcohol should be added to the reaction bottle. The concentration of sodium hydroxide is 40%. Chitosan with a deacetylation degree ranging from 15% to 45% is then added to the reaction bottle in a mass ratio of 1:4.8 with the sodium hydroxide solution.

Next, the pre-dissolved isopropyl ketone solution of chloroacetic acid should be slowly added to the reaction bottle while stirring. The stirring should continue for 3 hours after the addition is completed. The mass ratio of chitosan to chloroacetic acid is 5:1.

Once the reaction is completed, concentrated hydrochloric acid should be added to the reacted material in order to adjust the pH value to 6-7. The material should then be filtered with suction, and the filter cake should be washed with absolute ethanol three times. The washed filter cake should be dried below 50°C to obtain the carboxymethyl shell crude polysaccharide.
