4-Chlorobenzophenone Preparation

The main preparation methods of 4-chlorobenzophenone include Friedel-Crafts acylation reactions using various catalysts.

One common method involves the reaction of 4-chlorobenzoyl chloride and benzene under the catalysis of AlCl3. This method has a high yield but produces a significant amount of waste acid containing aluminum salts.

Another method is the condensation of benzoyl chloride and chlorobenzene under the catalysis of anhydrous AlCl3. While this method also yields 4-chlorobenzophenone efficiently, it requires a large amount of catalyst and results in the production of ortho-isomers.

A third method involves the use of a supported palladium catalyst for the cross-coupling reaction of tributylphenyltin and p-chlorobenzoyl chloride to produce 4-chlorobenzophenone with a high yield.

Lastly, a study reported the use of a catalytic amount of FeCl3/ZnCl2 as a catalyst for the Friedel-Crafts reaction of 4-chlorobenzoyl chloride and benzene under elevated temperature and pressure. This method allows for the efficient synthesis of 4-chlorobenzophenone with high purity.

4-Chlorobenzophenone Synthesis

Method 1: In a test tube, combine 4-chlorobenzoyl chloride, phenylboronic acid, and sodium carbonate. Flush the tube with argon and seal it with a rubber septum. Add toluene and water to the test tube. Transfer the reaction vessel to an oil bath maintained at 50°C and stir the mixture for 1 hour. Transfer the reaction mixture to a separatory funnel and dilute it with diethyl ether. Separate the aqueous phase and treat it with 3M HCl, 5% KOH, and brine. Dry the organic phase over anhydrous magnesium sulfate and pre-adsorb the crude product using chromatography grade silica gel. The product was purified by column chromatography on silica gel with ethyl acetate. Finally, evaporate to obtain the title compound 4-chlorobenzophenone.

Method 2: In a round-bottomed flask, add Na2CO3, Pd(OAc)2, [bmim][PF6], and H2O. Heat the solution to 60°C and add carboxylic anhydride and arylboronic acid. Stir the mixture at 60°C and then cool to room temperature. Extract the suspension with diethyl ether and combine the ether phases. Perform further purification by flash chromatography on a silica gel column. Wash the extracted residue with water in a recycling experiment. A second acylation reaction can be performed without additional Pd(OAc)2 or IL. Purify the compound using silica gel column. The final product obtained is 4-chlorobenzophenone.
